A Technique for the Simultaneous Dirt-free Lead Staining of Several Electron Microscope Grids of Thin Sections
نویسندگان
چکیده
The staining of thin sections of Osmium tetroxide (OsO4)-fixed biological materials with the lead hydroxide preparat ion of Watson (5) or with semisaturated solutions of lead subacetate (1) represents a considerable advance in electron microscope technique. The contrast of the image viewed in the electron microscope is so far enhanced that accurate focusing is now possible at appreciably higher magnifications. The contrast and detail visible in the micrograph also appears much improved, without the introduction of obvious artifacts. For routine work, a serious handicap is the formation of a precipitate of lead carbonate over a large proport ion of the section area. The method described by Peachey (4) considerably reduces this precipitate but allows only one grid to be stained per syringe. In the method to be described, as many as six grids can be stained simultaneously without any precipitation of lead carbonate.

on a conventional ultramicrotome. Semi-thin sections on grids were stained with the Giemsa method and methylene blue-azure II-basic fuchsin. Staining without removal of the embedding matrix was sharp and brilliant. These stained sections were observed under a light microscope. For electron microscopy, they were doubly stained with uranyl acetate and lead salt. Identical sites on such sections c...

To study the lead effect, we selected 14 females rats in which we had observed vaginal plague after mating, and divided them into a group of 8 female rats as the case group and 6 females as the control group. As drinking water the case group was given 0.13% lead acetate solution and the control group pure distilled water. Immediately after parturition the pups were killed and their livers remov...
متن کاملComparison of Three Staining Methods of Vibratome and Paraffin Sections in Preserving Thickness of Rat Hippocampal Sections
Purpose: To determine the optimal technique for maintaining hippocampus vibratome sections volume. Materials and Methods: This experimental study was performed on three rat hippocampus. The first hippocampus was embedded in paraffin and using rotary microtome forty sections were cut in a nominal thickness of 100 micrometer. They were then ordinary stained. The two others were cut into 80 sectio...
متن کاملNoncytopathic hepatitis A virus induces surface alterations in LLC-MK2 cells revealed by thin sections, negative staining, and scanning electron microscopy.
Previous electron microscope studies of ultrastructural events during hepatitis A virus replication in experimentally infected cells have used only ultrathin section techniques. Nevertheless, no important differences were observed between infected and uninfected cells. This study was carried out using scanning electron microscopy and negative staining of whole LLC-MK2 cells grown directly on gr...

The Boltzmann equation is a semiclassical approach to the calculation of the electrical conductivity. In this work we will first introduce a simple model for calculation of thin film resistivity and show that in an appropriate condition the resistivity of thin films depends on the electron mean free path, so that studying and measurement of thin films resistivity as a function of film thickne...
